ABSTRACT

This study aimed to determine the repellent and insecticidal activity of four essential oils (EOs) from plants collected in the Chocó rain forest, Colombia, against T. castaneum . Conventional hydrodistillation was used to obtain the EOs. The repellent and insecticidal activities were evaluated by the preference area and gas dispersion methods, espectively. Statistical differences (p<0.05) were determined by applying a student's t-test. EOs of Siparuna guianensis, S. conica, Piper marginatum, and Nectandra acutifolia showed excellent repellent properties as the main findings, highlighting S. conicaEO with 84% repellency (1-hµL/cm2), while P. marginatum showed to be bioactive to the dose of 500 µL/mL (72 h), inducing mortality of 100% of the exposed population. In conclusion, the results evidenced the repellent properties of the EOs evaluated against T. castaneum , which allows us to conclude that these plant species are potential natural sources producing bio-repellents that contribute to the integrated control of T. castaneum.

ABSTRACT

Asari Radix et Rhizoma (AR) is a traditional Chinese medicine with a history of more than 2 000 years of medication and has been included in ancient herbal works in the past dynasties. It is effective in releasing the exterior, dispersing cold, dispelling wind, relieving pain, opening orifices, warming the lung, and resolving fluids, and is still widely used in the clinical treatment of influenza, coronavirus disease-2019 (COVID-19) pneumonia, asthma, allergic rhinitis, eye pain, headache, toothache, oral ulcer, eczema, etc. Modern pharmacological studies have shown that AR has antipyretic, anti-inflammatory, analgesic, antibacterial, antiviral, relieving cough and asthma, anti-allergy, and other effects. AR contains a variety of chemical components, in which essential oil is not only associated with functions such as dispelling cold, relieving heat, relieving pain, and resisting inflammation and allergy, but is also toxic. AR also contains lignans, flavonoids, amides, phenanthrenes, alkaloids, and other non-volatile oil components, which play an important role in immunity regulation, anti-inflammation, pain relief, heart strengthening, and blood vessel expansion. The phenanthrene compounds are mainly aristolochic acid analogues, such as aristolochic acid Ⅳa and aristolochic lactam Ⅰ. Aristolochic acid Ⅳa has been proven to have a significant anti-inflammatory effect. The toxicity of AR is related to safrole, aristolochic acids and their analogues, and is also affected by many factors, such as preparation method, dosage, origin, collection time, medicinal part, and decocting time, which should be comprehensively considered in clinical application. Based on the relevant literature in China and abroad, the present study reviewed the correlation of chemical composition and pharmacological and toxicological effects of AR, and the safety of AR, aristolochic acid, safrole, and other components to provide a new perspective for an objective understanding of AR safety, as well as references for rational clinical application, production risk prevention and control, and drug scientific supervision of AR.

ABSTRACT

Abstract Oral candidiasis is a common fungal infection that affects the oral mucosa, and happens when Candida albicans interacts with bacteria in the oral microbiota, such as Streptococcus mutans, causing severe early childhood caries. C. albicans and S. mutans mixed biofilms are challenging to treat with conventional antimicrobial therapies, thus, new anti-infective drugs are required. Objective This study aimed to test a drug delivery system based on chitosan microparticles loaded with geranium and lemongrass essential oils to inhibit C. albicans and S. mutans mixed biofilms. Methodology Chitosan microparticles loaded with essential oils (CM-EOs) were obtained by spray-drying. Susceptibility of planktonic were performed according CLSI at 4 to 2,048 µg/mL. Mixed biofilms were incubated at 37ºC for 48 h and exposed to CM-EOs at 256 to 4,096 µg/mL. The antimicrobial effect was evaluated using the MTT assay, with biofilm architectural changes analyzed by scanning electron microscopy. RAW 264.7 cell was used to evaluate compound cytotoxicity. Results CM-EOs had better planktonic activity against C. albicans than S. mutans. All samples reduced the metabolic activity of mixed C. albicans and S. mutans biofilms, with encapsulated oils showing better activity than raw chitosan or oils. The microparticles reduced the biofilm on the slides. The essential oils showed cytotoxic effects against RAW 264.7 cells, but encapsulation into chitosan microparticles decreased their toxicity. Conclusion This study demonstrates that chitosan loaded with essential oils may provide an alternative method for treating diseases caused by C. albicans and S. mutans mixed biofilm, such as dental caries.

ABSTRACT

SUMMARY Introduction: Dental biofilm is a complex structure formed from the adhesion of various microbial cells to the film acquired on the dental structure. These cells adhere to each other while synthesizing an extracellular matrix, becoming a highly organized community, which makes it difficult to remove and promotes resistance to antimicrobials. The presence of Staphylococcus aureus in the oral biofilm can cause an disturb in the microbiota, facilitating the beginning of a disease process. Aim: To evaluate the non-adherent ability of Eucalyptus globulus and Eucalyptus citriodora against Staphylococcus aureus strains. Results: It was possible to observe that the essential oil of Eucalyptus globulus inhibited the formation of the biofilm with a Minimum Adhesive Inhibitory Concentration (MAIC) similar to 0.12 % chlorhexidine digluconate, both of 1:8. The essential oil of Eucalyptus citriodora did not prevent bacterial adherence to the tube walls in any evaluated concentration. Conclusion: Only the essential oil of the species Eucalyptus globulus has non-adherent activity against Staphylococcus aureus and can be used as an alternative product to control the biofilm.

ABSTRACT

The study is directed to establish the minimizing effects of Syzygium aromaticum, Ocimum sanctum, and Cananga odorata essential oils on the growth and ochratoxin A (OTA) level of Aspergillus ochraceus and Penicillium verrucosum in maize grains. S. aromaticum essential oil (SAEO), O. sanctum essential oil (OSEO), and C. odorata essential oil (COEO) were extracted by hydro-distillation technique, and a total of 50, 44, and 48 chemical constituents were identified by gas chromatography-mass spectrometry (GC-MS), respectively. The SAEO and OSEO belong to the chemotype of eugenol, whereas, COEO was found to be the chemotype of thymol, limonene, and ?-ylangene. The antifungal activity of essential oils (EOs) was determined by the micro-well dilution technique. The SAEO showed superior antifungal activity compared to OSEO, COEO, and synthetic antifungal agent nystatin, and its minimum inhibitory concentration (MIC) and minimum fungicidal concentration (MFC) values against A. ochraceous and P. verrucosum were noticed as 1251 ± 42.32 and 1878 ± 28.47 µg/mL, and 0815 ± 22.69 and 1146 ± 51.19 µg/mL, respectively. The antifungal mechanism of EOs was unveiled by assessing the intracellular reactive oxygen species (ROS), ergosterol content, and membrane integrity. The antifungal investigations found that EOs caused fungal mortality by increasing the intracellular ROS, depleting ergosterol synthesis, and distracting membrane integrity. Finally, antifungal and antimycotoxin activity of EOs was demonstrated in maize grains. The SAEO, OSEO, and COEO have reduced the complete fungal growth and OTA level of A. ochraceous and P. verrucosum correspondingly at 2500 and 2500, 3500 and 2500, and 3500 and 3500 µg/g in maize. The EOs could act as natural antifungal agents; protect foodstuffs from fungal infection and mycotoxins during storage.

ABSTRACT

Basil (Ocimum basilicumL.) is a medicinal species used in several areas, such as food, medicines and cosmetics, and the understanding of its physiological behavior under environmental conditions is of paramount importance for the improvement of cultivation methods. The objective of this study was to evaluate the influence of different water availability under physiological, biochemical and metabolic characteristics, in three distinct genotypes: 'Alfavaca basilicão', 'Gennaro de menta' and 'Grecco à palla', during two different phenological stages (vegetative and reproductive). It was found that the water deficit promotes physiological changes to tolerate water stress, and the studied genotypes have different routes to achieve this physiological tolerance, which culminates in a distinct accumulation of metabolites in plants, and can be considered interesting if the final product is the production of essential oils.

ABSTRACT

One of the main groups of preservatives used in most cosmetics are parabens, that many studies have shown that they can cause allergic reactions such as contact dermatitis, among other skin sensitizations. To minimize these problems, the industry is producing cosmetics preservative free or using natural products instead and their combination with the synthetics. Among the natural preservatives used, we can mention essential oils as a viable alternative. In this context, the present work aims to experimentally evaluate the antimicrobial potential of the Conobea scoparioides Cham. & Schltdl. essential oil, popularly known as pataqueira, the effect of its association with parabens and its effectiveness as a preservative in cosmetic bases. The essential oil composition was analyzed, indicating that it is composed mostly of terpenes, with thymol methyl ether (39.2%), thymol (33.8%) and -phelandrene (15.9%) as major compounds. The antimicrobial activity of essential oil and thymol was accessed through the minimum inhibitory concentration (MIC), whose results in µg/mL for essential oil and thymol were respectively: Staphylococcus aureus 650.70 and 284.90, Escherichia coli 721, 53 and 271.20, Pseudomonas aeruginosa 1748.00 and > 2,000, Burkholderia cepacia 833.03 and 1,077.70, Candida albicans 521.43 and 172.61 and Aspergillus brasiliensis 300 and 400. The synergistic effect of the association of essential oil with parabens was performed through a centroid simplex experimental design for a mixture of methylparaben, propylparaben and essential oil against the same microorganisms used in the antimicrobial activity evaluation The ideal concentrations obtained by statistical analysis for each component in µg/mL were: 1120 for methylparaben, 350 for propylparaben and 675 for essential oil. The effectiveness test of the preservative system in cosmetic formulation was carried out using the ideal concentrations plus two higher concentrations and one below the ideal. For all challenged microbial strains, the test result was a total reduction of the inoculated microbial load in the seven days of testing and no increase until the twenty-eighth day, which demonstrates the effectiveness of the association of essential oil with synthetic preservatives. C. scoparioides essential oil showed an important antimicrobial potential both alone and in association with parabens. These results demonstrated that it can be used to compose a preservative system for cosmetic formulations containing lower amounts of synthetics

ABSTRACT

Aims@#This study evaluated in vitro resistance and susceptibility of Enterobacteriaceae (Escherichia coli and Klebsiella oxytoca strains) and Staphylococci strains, isolated from sheep’s milk, against antibiotics and essential oils from Thymus satureioides and Mentha pulegium.@*Methodology and results@#Antibiotic resistance tests were done using disc diffusion while essential oils were extracted by steam distillation and yields were calculated relative to plant dry matter. Gas chromatography-mass spectrometry (GC-MS) was used to analyze each oil's chemical composition. Amoxicillin + Clavulanic acid (AMC), Cefotaxime (CTX), Cefoxitine (FOX), Nalidixic acid (NA), Gentamicin (CN), Ciprofloxacin (CIP) and Ofloxacin (OFX) were very effective against the E. coli strains tested. Half of the strains were resistant to AMC, 60% to Ticarcillin (TIC) and 80% to Tetracycline (TE). Klebsiella oxytoca was resistant against AMC, FOX and TIC (100%). Antibiotic-resistant testing on Staphylococci strains indicated Staphylococcus capitis and S. chromogenes as the most sensitive. Staphylococcus aureus, S. xylosus and S. cohnii ureal exhibited less resistance to Oxacilin (OX), TE, Pristinamycin (PT), Erythromycin (E) and Penicillin (P). Mentha pulegium resulted in a higher yield of essential oil of 3.2% oil compared to T. satureioides with only 1.85% yield. The monoterpene oxygenated derivatives, monoterpene hydrocarbons and phenols are found in essential oil extracts. Thymus satureioides essential oil had high antibacterial activity even at low concentrations (0.2; 0.55 g/mL). The minimal bactericidal concentration (MBC) values indicate that the essential oils from the plants analyzed had bactericidal effects on all strains tested and are similar to the minimal inhibitory concentration (MIC) values.@*Conclusion, significance and impact of study@#The high antibacterial properties of these medicinal plants, against bacteria isolated from sheep’s milk, provide an opportunity to use these medicinal plants in the breeding sector, as additives and preservatives in the dairy industry.

ABSTRACT

Dysmenorrhea is a common gynecological disease in clinic, with primary dysmenorrhea and secondary dysmenorrhea. Primary dysmenorrhea is usually not accompanied by organic lesions in genital organs, which is mainly related to the increase of prostaglandin content in endometrium during menstruation. Secondary dysmenorrhea is accompanied by organic lesions of reproductive organs, often associated with local lesions of reproductive organs, but also with patients' mental factors and neuroendocrine factors. The incidence of dysmenorrhea is as high as 73.8%, and there is no radical cure method, which has a great impact on the life, work and learning of patients. Chinese medicine essential oil widely exists in aromatic Chinese medicine, with antibacterial, antioxidant and anticancer activities. It can regulate neuroendocrine function, anti-inflammatory and analgesic effects, and improve mood by regulating the levels of prostaglandins, oxytocin and other hormones in the body and regulating the emotions of patients, there by alleviating dysmenorrhea to a certain extent. In recent years, many scholars have made more in-depth research on Chinese medicine essential oil in alleviating dysmenorrhea, but there is a lack of comprehensive collation of such studies. In this regard, the author has systematically sorted out the generation and classification of dysmenorrhea, the mechanism of action of essential oil of traditional Chinese medicine to alleviate dysmenorrhea and the application of essential oil of traditional Chinese medicine in the field of dysmenorrhea by consulting relevant literature in Chinese and foreign languages in recent years, so as to provide reference for the treatment of dysmenorrhea.

ABSTRACT

Essential oils(EOs) from Chinese medicinals, which can be used as adjuvants and exert certain therapeutic effect, are directly used in Chinese medicine formulas. Conventional research strategy for EOs from Chinese medicinals is to compare the efficacy of the prescriptions before and after the addition of EOs, and the penetration-enhancing mechanisms of EOs remain unclear. In modern research on EOs from Chinese medicinals, the method for studying chemical penetration enhancers is often used, which fails to reflect the overall efficacy of EOs. This study clarified the property regularity of EOs from Chinese medicinals as transdermal penetration enhancers, and thereby proposed a research model which integrated the medicinal and adjuvant properties of EOs from Chinese medicinals via "component-delivery-effect" characterization route. The core concept is that constituents of EOs from Chinese medicinals and their delivery process play a key role in their external application. This research model is expected to serve as a reference for further research on EOs from Chinese medicinals for transdermal application.

ABSTRACT

Essential oils from Chinese medicine are popular in the fields such as medicine, food, and cosmetics because of their unique biological characteristics. However, since essential oils are lipophilic compounds with high volatility, poor stability, and strong irritation, various preparation technologies need to be employed to improve stability, reduce irritation, and increase bioavailability. At present, a variety of preparation technologies have been applied to the encapsulation of essential oils. Various encapsulation strategies are formed because of different delivery systems featured with multiple principles and characteristics and are widely used to improve the stability of essential oils. Essential oils of Chinese medicine are widely used in the medical field, and they are under continuous innovation and development in clinical research, the pharmaceutical industry, medical products, etc. The present study summarized various delivery systems that could improve the stability of essential oils and reviewed the applications of essential oils encapsulated in the delivery systems in the medical field to provide re-ferences for the improvement of stability of essential oils and their safety, efficiency, and wide use in the medical field.

ABSTRACT

The utilization of antimicrobials in animal production, causes selection of resistant bacteria. The objective of this study was to compare the utilization of alternatives in association with preventive antibiotic therapy in swine feed during the growing and finishing phases. 1,045 animals were used from 60 to 190 days of age and were subjected to six treatments with 16 repetitions as follows: 1) antibiotic free; 2) antibiotics; 3) prebiotic; 4) probiotic; 5) essential oils; and 6) organic acid. Animals were weighted, and clinical history was recorded including mortality and diarrhea. At the abattoir, pneumonia index and gastric ulcers were investigated. The cost for each treatment was discussed. No difference between treatments were observed (P>0.05) regarding feed conversion rate (2.64±0.03), overall average weight gain (107.06±0.9kg), average daily weight gain (856.49±7.7g) and carcass weight (92.4±0.7kg). The application injectable drugs in animals presenting clinical symptoms, represented US$ 0.56/intervention, without difference between the treatments (P>0.05). Furthermore, independently of the treatment, high frequency of pneumonia was observed (>0.90). No difference for the degree of gastric ulcer nor feces consistency were observed (P>0.05). The utilization of antibiotic therapy and alternatives to antibiotics in feed did not produce benefits to the production indices and sanitary performances of the animals.(AU)

ABSTRACT

ABSTRACT The aim of the present study was to analyze the chemical composition ofCurcuma longa,Pimenta dioica,Rosmarinus officinalis, andSyzygiumaromaticumessential oils (EOs) and their antifungal and anti-conidiogenic activity againstFusarium verticillioides. The chemical profile of the EOs was determined by GC/MS. The antifungal and anti-conidiogenic activities were evaluated by the agar dilution method. The tested concentrations were 1000ppm, 500ppm, 250ppm and 125ppm.S. aromaticumEO exhibited the highest antifungal effect, followed byP. dioicaand to a lesser extentC. longa. The major compounds of these EOs were eugenol (88.70% inS. aromaticumand 16.70% inP. dioica), methyl eugenol (53.09% inP. dioica), and α-turmerone (44.70%), β-turmerone (20.67%), and Ar-turmerone (17.27%) inC. longa.Rosmarinus officinalispoorly inhibited fungal growth; however, it was the only EO that inhibited conidial production, with its major components being 1,8-cineole (53.48%), α-pinene (15.65%), and (−)-camphor (9.57%). Our results showed that some compounds are capable of decreasing mycelial growth without affecting sporulation, and vice versa. However, not all the compounds of an EO are responsible for its bioactivity. In the present work, we were able to identify different major compounds or mixtures of major compounds that were responsible for antifungal and anti-conidiogenic effects. Further experiments combining these pure components are necessary in order to achieve a highly bioactive natural formulation against the phytopathogenic fungusF. verticillioides.
